It mostly has to do with people expecting there to be a joke, and the whole thing being such a non-sequitur that their brains hurt themselves and they wind up assuming "honk honk" IS a joke, as far as I can tell.
I've heard, however, some lines about how "the stupider the YTMND was to begin with, the funnier it is when it becomes a meme." It's basically just that something so stupid becomes funny as a totally nonsensical reference.
