I don't know that there's necessarily a punchline here.
If you look at the source, this was part of a manga sample for a book being sold at comiket. I don't think this page was meant to stand alone.

Tsubasa's Koizumi is straight as an arrow and without any other context besides the SFX, it seems that since Kyon is gawking at Mikuru again, Koizumi is speaking to him threateningly.

See, my interpretation is that he was trying to hook Kyon up with Haruhi. Threatening isn't really Koizumi's style, especially in this artist's series. Trolling certainly, but not threatening.

Although I still think we would need to see the next page/rest of the book to know what's going on for sure.
