I could be wrong, because I'm not 100% sure what the symbol in the fifth panel (u) translates into an "uhh..." sound (one of the translations I got was "help"). However, iya, while meaning no, could also mean yes, as well as saying "well" ie. maybe.

The response in panel five is more a choked grunt. The "Iya" in panel 6 is Reimu speaking, and in context with the rest of her dialog afterward, makes sense as "no" here.
